Capsule material is a formulation variable and not the packaging. The material of the shell determines the amount of moisture and oxygen which reach the contents, the speed at which the capsule dissolves, and how long the active ingredient remains stable. The three shell materials used in the manufacture of supplements are gelatin, HPMC and pullulan, each of which behaves differently when exposed to moisture, oxygen and dissolution conditions.

Gelatin shells hold 13-16% equilibrium moisture. In dry conditions, gelatin releases moisture into hygroscopic fills, degrading them faster. In humid conditions, gelatin capsules absorb moisture, softening the shell and speeding degradation of moisture-sensitive actives.

HPMC vegetable capsules hold 2-8% moisture and resist transfer in both directions, making HPMC the standard choice for moisture-sensitive actives and high-humidity markets.

HPMC is the preferred shell for moisture-sensitive actives, due to low equilibrium moisture and reduced transfer. Hygroscopic actives such as magnesium salts, choline bitartrate, vitamin C, creatine monohydrate, some amino acid blends, absorb moisture from the shell, causing clumping, caking, potency loss, or premature dissolution. For these actives, a low-moisture shell is a stability requirement, not an upgrade.

When should pullulan be used for oxygen-sensitive actives?

Pullulan should be used when the active degrades by oxidation. Pullulan empty capsules are fermented from tapioca starch and permeate less oxygen than gelatin or standard HPMC, extending potency for CoQ10, omega-3s, beta-carotene, lutein, astaxanthin, and probiotics. Pullulan is also plant-derived, non-GMO, and free of BSE/TSE, supporting clean-label formulations.

Shell quality determines how predictable shelf life will be. Variation in moisture content, wall thickness, or polymer consistency produces inconsistent stability results across batches, even with an unchanged formula. 

Fill weight compounds this: excess headspace after filling raises the oxygen volume in contact with the fill, accelerating oxidative degradation, so capsule size should be matched to fill weight using the empty capsule size chart, not chosen for dosing convenience alone.

  • Gelatin becomes brittle and cracks below ~12% relative humidity.

  • Discoloration or spotting signals a chemical reaction between shell, fill, or packaging.
  • Delayed or incomplete dissolution occurs when gelatin is exposed to aldehydes from certain fill ingredients.
  • Clumping or potency loss occurs when moisture migrates from a gelatin shell into a hygroscopic active.
